TALEN™ Sure KO

Highest knock-out success

TALEN™ Sure KO binds its recognition site within the first coding exon, inducing a DNA double-strand break at the recognition site which is repaired by error prone non homologous end-joining (NHEJ).

The introduction of frame shift mutations in the start codon region of a gene avoids the production of partially functional truncated proteins and greatly ensures gene knock-out events.

The very high modularity of TALEN™ engineering guarantees that there is always a TALEN™ Sure KO for any gene knock-out you want to achieve.

TALEN™ Sure KO’s activity is validated at the level of your choice (on episomal or chromosal target sequence). Click here to see the different validate levels.

    In-house results

Internal data demonstrate that TALEN™ Sure KO efficiently knock-out a GFP transgene that has been stably integrated as a single copy in the CHO-K1 genome, with efficiencies of up to 87% within 6 days post TALEN™ transfection.

    (A) CHO-K1 cell line stably expressing a single copy of the GFP gene (red curve), compared to a GFP negative CHO-K1 control cell line (blue curve) was transiently transfected with GFP specific TALEN™ Sure KO expression vectors; (C) 6 days post transfection, 87% of cells were found GFP negative (red curve); (B) and (D) confocal images corresponding to (A) and (C) respectively.
