cGPS® Targeted Integration is a breakthrough in stable isogenic cell line generation: easy- and ready-to-use, saves time, effort and money!

With cGPS® Targeted Integration kits, sequence-specific nuclease-driven targeted integration can easily be achieved, it requires only the presence of a nuclease recognition site within the targeted cell genome. Although a natural nuclease recognition site is not present in mammalian cells (since the used nucleases are originating single-celled organisms), we propose two ways to carry out sequence-specific targeted integration in higher eukaryotic cells:

  1. An engineered cell line is carrying the recognition site of a natural meganuclease as a single copy. This is the principle of our cGPS® line of Targeted Integration kits;

  1. An engineered nuclease targets a native, endogenous genomic locus (e.g. a safe harbor). This is the principle of our cGPS® Custom line of Targeted Integration kits.

The key benefits of cGPS® sequence-specific nuclease-driven targeted integration are:

  • Easy and ready to use:
    • cGPS® TI kits come with an optimized user manual, which allow an easy set-up of you experiment;
  • Fast and time-saving:
  • Targeted cell population can be recovered in as little as 2 weeks, isolated clones in 3 to 4, due to the use of selection agents
  • Only little clone screening is needed (use of selection agents)
  • High clone homogeneity is obtained thanks to very efficient targeted integration protocols (95%+ targeted integration)
    • cGPS® TI products are immediately retargetable cell platforms. There is no prerequisite to first establish such a retargetable platform. Isogenic cell line generation is therefore very fast.
  • Effortless, trusted results;
  • Trusted results saving time and money:
    • Guaranteed and predictable transgene expression: integration is performed in a single selected transcriptionally active genomic site;
    • Robust data comparison: the generated clones are isogenic;
    • Highly stable expression: for at least 20 weeks, even in the absence of selection agents;
    • Reproducible, highly efficient integration of a single transgene copy at a unique genomic site.
